William's parents moved to Lewis County, West Virginia (then Virginia) by the time of the 1850 census. William was still residing in Nelson County with some of his mother's siblings.
William and Elizabeth Ann Wingfield were married September 23, 1852 in Lovington, Nelson, Virginia. They were the parents of twelve children:
An unnamed son born in 1853 who only lived a few days, Eliza Lucinda, Robert Alexander, Mary Frances "Mollie", John Hiter, Amanda J, Margaret Susan, Mildred Mae, William Henry, Martha Emmaline, Edward Morgan and Lillie Josephine.
The family moved from Nelson County Virginia to Augusta County Virginia and then to Braxton County, Virginia where they settled on Dumpling Run Ridge near Burnsville.
William was a farmer.
The following is an excerpt from Hardesty's West Virginia Biographical Atlas published in 1882:
"William W. Henderson, son of Robert C. and Lucinda (Lobban) Henderson, was born in Nelson County, Virginia, September 30, 1825. In Staunton, Virginia, July 16, 1861, he enlisted in Company 1, 62d Virginia Infantry, and with that company he took part in the battles of New Market, and those fought about Richmond, where for three days he was constantly in skirmishes, and the battle of Whitehouse. After this engagement he was stricken with fever and taken home, and before his recovery the war was ended."
