The son of Martin and Anna Katherine (Metts) Souter, Jacob was born in Newberry County, SC. Of German extraction, his family had settled in the Newberry area during a time in which many Germans were settling there, and the German region became known as the Dutch Fork. His family surnames were almost certainly originally "Sauter" and "Metz" before being Anglicized.

Jacob served in the War of 1812 and Creek War as a private in Nathaniel Martin's Company, Nash's South Carolina Regiment. After the war, he married Rachel Vardaman on January 24, 1815. Rachel was partially of German extraction herself, although it was Low German in contrast to Jacob's High German ancestry, and she also had English and Dutch ancestors.

Jacob and Rachel raised a very large family in South Carolina of 14 children, before moving as a family to Pontotoc County, Mississippi. There, they became members of the Cherry Creek Baptist Church. Rachel died in 1854, and Jacob followed in 1880. They were both buried in the Cherry Creek Cemetery.
