John was born in Howard Twp., Centre Co., PA, the sixth child of John Grube and Barbara Hoy. He married Louisa Weiss on March 10, 1853, presumably in Jefferson Co., PA. He died on the home farm in Bell Twp., Jefferson Co., PA.

John's parents moved from Centre County to Jefferson County in 1833, purchasing a large tract in Young Twp. (now Bell Twp.), which was later divided up among the Grube sons.

John and Louisa were the parents of thirteen children:
1. Joseph Irwin Grube
2. "Jane" Martha Jane (Grube) Depp
3. Henrietta (Grube) Depp
4. "Albert" Henry Albert Grube
5. Jacob Wright Grube
6. "Means" Augustus Means Grube
7. Margaret Matilda (Grube) Brown
