After the Civil War he founded a pharmacy in Richmond where he developed medicines for domestic amimals. He turned that into a successful company he named Sergeant's after his dog. Sergeant's remains a leading pet food and medicine company. Miller learned to play the banjo as a child and was noted for his talent at a young age. In the 1890's, after turning over his business to his son, he became a full time performer.

Recordings of Polk Miller and the Old South Quartette were released on Edison cylinders. Nearly a hundred years later in 2008 some of these recordings were transferred to a CD.
