Born in Milford, New York, to John, Sr., and Mary (Loughead) Moore. John's father was a prominent citizen who served in the state assembly. John's mother died when he was 6 years old, and his father remarried a widow with 6 children. John was this raised in a large blended family with his sister, step-siblings, and three new half-siblings.

Married Anna Maria Starr at St. Paul's Episcopal Church in her hometown of Brookfield, Connecticut, on October 7, 1830. Together they had two children:
1. Robert Sayles Moore (b. December 22, 1833, near Lebanon, Ohio)
2. Cornelia Maria Moore (b. December 28, 1837, near Union Mills, Indiana)

John co-owned a mercantile business in Milford with his step-brother, Alonzo Bridges. After his marriage, John sold his share of the business and moved west with his wife and parents. They eventually settled near Union Mills, where they were frontier farmers. Despite his old age, John enlisted as a private in Company G of the 29th Indiana Infantry in February of 1864. He served for a year before mustering out in Marrietta, Georgia, and would die later that year at the age of 61.
