Bishop Agostino Zampini OSA., of the Titular See of Porfireone, was born on August 22, 1858, in Florence, Italy, and joined the Order of the Augustinian Fathers at an early age. He received the Augustinian habit at the convent of Santo Spirito of Florence on February 2, 1880, took his vows on February 12, 1881, and was ordained to the priesthood on April 12, 1884.

Forming part of the community of Santa Maria in Selva di Borgo at Buggiano between 1884 and 1894, he was assigned to Santa Monica in Rome, serving as secretary of the Order between 1895 and 1901, assistant general from 1905 till 1907, postulator general and prior of Sant'Agostino. As postulator he took care among others of the causes of Blessed Stefano Bellesini OSA., and Saint Rita of Cascia. Elected Minister Provincial of Toscana, he moved once again to the Santo Spirito convent of Florence.

On December 9, 1910, Zampini received a telegram which informed him of his appointment as Vicar General for the Vatican City State or more formally the Vicar General of His Holiness for the State of Vatican City dealing with the spiritual needs of the Vatican City, similar to the post of Vicar General of Rome that deals with the day to day administration of the Diocese of Rome.

Zampini received his episcopal consecration with the Titular See of Porfireone on January 6, 1911, from Cardinal Rafael Merry del Val y Zulueta, Secretary of State of the Vatican City, at Sant'Agostino in Rome.

During his 26 years of episcopate, as Sacristan of His Holiness, Msgr. Zampini served as principal co–consecrator in the episcopal consecrations of Cardinal Gaetano De Lai, Cardinal Adam Stefano Sapieha, Archbishop Louis Petit, Bishop Ramón Zubieta y Les OP., Cardinal Pietro Fumasoni Biondi, Eugenio Maria Giuseppe Giovanni Pacelli - future Pope Pius XII - Cardinal Francesco Borgongini Duca and Bishop Kidanè-Maryam Cassà.

Monsignor Zampini died at the age of 78, on June 7, 1937, at the Apostolic Palace of the Vatican City.
