A daughter of James Alexander and Margaret Clark Alexander, Agness was born in Scotland and came with her parents to the U.S. They settled in York, Pennsylvania, where Agness met and married William Pickens on 5 Jun 1789. The entire family (the Alexanders, Clarks, Pickens and other related Scots families) then moved to Belmont Co., Ohio and settled in the area now known as Scotch Ridge.

Agness and William Pickens had 9 children: Margaret, Thomas, James C., Ellen or Eleanor, William, Janet or Jeanette Jane, Nancy Alexander, John Clark, and Alexander.
