Jacobus was the son of Jacobus Stryker and Jannetje Van der Beek of Flatbush, Long Island and New Brunswick, New Jersey. He was married about 1747 to Sarah Messler, the daughter of Johannes and Catharine Metselaer of Middlebush, New Jersey. Jacobus and Sarah had five known children:

* Elizabeth, b. c. 1748 (plus or minus 2-3 years; her mother was born in 1728 and her son Archibald was b. 1768, making the window for Elizabeth's birth fairly narrow). She married, as his second wife, Archibald Thompson, who had first married Coba Shureman. Elizabeth had two children: Archibald, Jr (b. 1768 and buried in New Brunswick, NJ) and Jane.

* Catrentje, b. say 1752

* Janitje, baptized at the New Brunswick DRC 14 September 1755, m. Francis Cowenhoven.

* Johannes, baptized at the New Brunswick DRC 15 November 1758, d. in New York in 1783, and married 10 July 1779 Elizabeth Stryker.

* Rev. Peter, b. 23 December 1763, m. Sarah Barkeloo.
