Isaac Bush was baptized on 2 March 1718 in Manhattan, a son of Justus Bush and Anne Smith. His baptismal sponsors were his siblings Justus Bush, Junior and Charity Bush.

He was mentioned in the wills of his father (24 June 1737) and mother (6 October 1745).

On 10 September 1746, he was one of the witnesses for the will of his brother Justus Bush of Horseneck in Greenwich, Fairfield County, Connecticut, and on 2 December 1760 he affirmed his presence as a witness at the will's probate.

On 20 March 1768, not long after his fiftieth birthday, Isaac Bush of Greenwich journeyed together with James Mosher and Abel Mosher to the Manor of Queens Village on Lloyd's Neck, Long Island, New York, located about 5 miles southeast of Greenwich across Long Island Sound. All three men confessed to trespass on 9 April 1768 and were fined for "Digging of Clams, Repairing an old house & Hunting game with guns".

On 7 February 1784, Isaac Bush and his wife Elizabeth sold their house and homelot in Greenwich to his nephew and neighbor Dr. William Bush, together with their canoe lying in Horseneck Harbor. The house was situated on present-day Field Point Road near the bridge crossing Horseneck Brook and near the head of Bush's Harbor (present-day Greenwich Harbor).

On 10 May 1792, Isaac Bush was listed as a "sufferer" of the Town of Greenwich, having sustained property damage during the Revolutionary War (1775–1783).

Isaac Bush was presumably buried in the family cemetery (Bush Family Burial Ground at Horseneck). Unidentified remains from the Bush Vault at Horseneck were removed to the Bush plot in Putnam Cemetery in 1894.
