Layne was the son of Hiram Compton and Janney Shannon. He married Melsena Hatton, Mary McDowell, Louisa "Vicie" Miller, and Abigail Little.

Layne was residing in Rockwell, KY, when his first wife Melsena died in childbirth after a year of marriage. He returned to Blaine, Lawrence Co., KY with his infant son, James Hatton Compton. In 1850, Layne was a raftsman on the Big Sandy River, along with his brother-in-law, Melville Hatton. Another brother-in-law, Hickman Davis, was his neighbor and a river pilot on the Big Sandy. He became a farmer, raised a large family, and was a church deacon. Layne died when struck by a moving wagon in Louisa, at the age of 72.
